The event was scheduled to run on March 23 at the Halls Gap Centenary Hall but has been cancelled due to a lack of ticket sales.
Event organiser Angeline Martin said small events such as the comedy festival need community support.
"This year we just couldn't sell the tickets unfortunately," she said.
"You need a lot of support in a small community like this but even with last minute ticket sales we wouldn't have sold enough to fill the hall.
"Sometimes that's just how it goes. It's a bit of a pity because it was such a good event in the past."
Australian comedian Geraldine Hickey and international acts Viggo Venn and Zach Zucker were scheduled to perform at this year's festival.
The festival was successful in the previous two years but was unable to hit the mark this year.
Ms Martin said she believes there is still a place for comedy in Halls Gap.
"There are a few people in town who want to push the idea of comedy, I think it will work but maybe we need to look at doing things differently," she said.
"The acts love coming to Halls Gap. The event is over for this year but definitely not done for good.
"Maybe we change it to a series of smaller events rather than one big festival."
Grampians Tourism chief executive Marc Sleeman said he hopes to see the event return next year.
"It's disappointing to see it not run this year since in the past it has been a great event," he said.
"We look forward to supporting it to come back in 2020."
Ms Martin said while it's disappointing to miss out on the festival this year, there are plenty of positives for the town.
"There are always a lot of great things happening in Halls Gap so it's not like this is the end of the world," she said.
"A key thing we can take from this is community events need support in anyway, whether that be doing things like sharing on Facebook or putting a poster up."
