The Cats have appointed Mackie, who played 280 games for the club, as their list manager.
Wells will stay in charge of recruiting.
Wells is one of the AFL's top recruiting gurus and previously had held the two roles at Geelong.
He briefly left the club earlier this year, but returned and continued to run their list management and recruiting.
Cats football boss Simon Lloyd said Mackie assumed more list management duties during the season.
"We all believe he is ready to take that role on a full-time basis," Lloyd said.
"At the same time, we are really pleased that Stephen will continue as our recruiting manager. He continues to be at the top of his profession."
