The driver of the truck, 20-year-old Dandenong North man, has been airlifted to hospital with life-threatening injuries.
A second driver, a 64-year-old Wangaratta man, has been transported by road to hospital with non-life-threatening injuries.
The third driver, a 61-year-old Bayswater man, was taken to hospital for observations.

The Western Highway is still closed after a multiple truck collision wreaked havoc at 1am on Friday, April 16.
A CFA spokesperson said three B-double trucks collided earlier this morning. CFA, SES and Victoria Police were on scene.
One driver was trapped and required extrication.
A police spokesperson said the driver of the truck has been airlifted to hospital with life-threatening injuries.
A second driver has been transported by road to hospital with non-life-threatening injuries.
The third driver was taken to hospital for observations.
IN OTHER NEWS:
One vehicle was a refrigerated truck carrying over 200 pig carcasses which scattered across the area, the CFA spokesperson said.
It is expected that the Western Highway will remain closed for some time while investigators remain on the scene.
Another vehicles was carrying potatoes and the other was general freight. Two trucks are still on the scene.
Western Highway is closed between Nhill and Dimboola due to the crash and clean up job.
Adelaide bound vehicles are to divert from Dimboola to Jeparit and then to Nhill. Melbourne bound vehicles are to detour from Nhill to Jeparit and then to Dimboola.
Emergency services are on site and a signed detour is in place.
The exact cause of the collision is yet to be established and the investigation remains ongoing. the police spokesperson said.
