Nhill will host the prestigious battle between the two states for the fourth time in 15 years.
Victoria Country assistant manager and Nhill Golf Club greenskeeper Shane Grover said it was shaping up as a big weekend for the region.
"It's a big honour for a little country town to get something like this," Grover said.
"It's a pretty prestigious event which has been going for a long time, so it's a pretty big event for our golf club.
"I've been telling our members and everyone else that it is really worth watching. It's really incredible golf these guys play, so hopefully they can get around it, especially during the singles match-play on Sunday."
Ten of the best players from country Victoria and South Australia have been selected to compete.
Two Wimmera-based players have been selected in the Victorian side, with Stawell's Tim Coffey representing his club Taralgon and Adam Tribe representing Halls Gap and Chalambar.
Coffey is the reigning Wimmera champion and represented Victoria Country at the 2018 event alongside Grover when it was hosted in Mount Gambier.
The two were part of a Victorian team that claimed victory and seized back the country shield after losing the previous two iterations of the event.
Coffey won all three of his games last year.
In addition, for the first time in the event's history, two female players have been included from each state as part of the ten-person team.
Teams will tee off at 8AM on Saturday and Sunday. Saturday morning will be foursomes match-play, while in the afternoon the competition is 4-ball-best-ball match play. Sunday will be singles match-play.
Grover was hoping the course would be in top condition, despite incredibly hot conditions mid-week on Wednesday and Thursday.
"I'm really happy with the condition of the course," Grover said.
"It's a really great chance for us to showcase our course to some top-notch golfers."
