For the second year, the Giants and Warriors will do battle for the Grampians Excavations Cup, with the winner to take home $5000.
"It's a big day for both clubs, and we are privileged to have the opportunity to compete for something like this once again," Stawell Warriors president Tim Williams said.
The Warriors are yet to record a win over the Giants from their three meetings since last year, including in the corresponding cup game in 2018.
Stawell coach Damian Joiner said there is plenty on the line.
"There is that extra incentive to get that maiden win over them as well as top off a big day for the club with the prize on the line," he said.
"It really is an eight point game with them a game behind us, it is massive."
With all teams having played each other once, marking the halfway point of the season, Stawell sits in third spot, one win clear of the fifth-placed Giants.
The Giants got the job done against Stawell in round one, but the Warriors go into the game coming off the back of three consecutive wins.
"We would have liked to be a few more wins up at this time of year but we will take where we are at right now," Joiner said.
"It is important not to get complacent. I think it is the perfect time to play a team like the GIants. We are playing good footy, our confidence is high and they will be a big test for us."
The Warriors are still waiting on the fitness of 2017 best and fairest winner Sean Mantell, who has managed just two games this year.
"Sean is nearly right to go, in the next few weeks he should be ready," Joiner said.
Things are also looking positive on the netball court, with the Warriors coming off the back of their first win of the season.
Stawell co-coach Toni Stewart said it is important to stay grounded after a season-high last weekend.
"It is good to have that winning feeling again but there are still aspects of our game we have been working on," she said.
"I think a big thing we need to improve on is recognising when we are falling in a hole and regrouping on the court to ensure we don't fade out."
Between 15 and 25 millimetres of rain is expected to fall in Stawell on Saturday, which Stewart said will have to be accounted for.
"We haven't played well when it has been wet so far this year so we will need to really focus and cherish the ball," she said.
